Von Kossa Gieson staining

Standard staining for acrylate

Important: Do not use metallic objects for this staining!

·        Deplasticizer I - III              each 5 min.                              

·        2´Ethanol 100%                  each 2 min.                              

·        Ethanol 96%                       2 min.

·        Ethanol 80%                       2 min.

·        Ethanol 70%                       2 min.

·        Ethanol 50%                       2 min.

·        Aqua dest                           wash by submerging 3-5 times

·        Silver nitrate 3%                 5 min.                          

·        Tap water                           rinse for 10 min.

·        Soda-formol                        5 min.

·        Tap water                           rinse for 10 min.

·        Sodium thiosulfat                5 min.

·        Tap water                           rinse for 10 min.

·        Van Gieson-stain                20 min.

·        Tap water                           rinse shortly

·        Ethanol 80%                       rinse shortly

·        Ethanol 96%                       rinse shortly

·        2´Ethanol 100%                  rinse shortly

·        Xylol I - III                          each 5 min.

·        Mount with DPx:                 Drip DPx with a glass rod onto the cover slip and place it on the section  

Solutions:

3% Silver nitrate.

3g Silver nitrate per 100ml aqua dest.

Soda-formol

12,5g Sodium carbonate

187ml aqua dest.

62,5ml 37% Formaldehyde

5% Sodium thiosulfate

5g per 100ml auqa dest.

van Gieson stain

2,5g acid fuchsine

900ml saturated picric acid

100ml glycerol

5ml concentrated nitric acid

Staining results:

black:               mineralized bone

red:                  Collagen and connective tissue

yellow:              muscles and erythrocytes
